These are two dofferent outputs with sperate settings and different functionalities.
If you monitor via a guitar combo you should use the MONITOR OUT, feed the signal into the effect return of the guitar amp and activate Monitor Cab Off, which only works for the MONITOR OUT. Under no circumnstances you should feed a signal into the guitar input of a guitar amp..

It will color the sound of the Profile since you are introducing the Pre Amp tone of the Tweed along with the profiled Pre Amp of the Kemper. It can sound "good" but is not accurate. Plugging into the Tweed FX loop will bypass it's Pre Amp giving a more accurate sound of the profile you have loaded. I think you can also overload the Tweed Pre Amp using Monitor Out on it. Direct Out set at Git Analog is for Pre Amp input.

It might sound "fine" but it won't be as good as the Kemper is capable of. There are lots of threads on here explaining why plugging a Kemper into the input of a tube amp reduces the quality of both tools.

The MONITOR OUT is feeding a line level signal. Your Fender's guitar input is instrument level. The signal should be way too hot and the whole setup should be very noisy. This is no recommended setup.

You can't use guitar amplifiers without effect loop as amplifiers for the PROFILER.
Even if the guitar amplifier has an effect loop, we are not recommending such a setup, because normally - even if you suppress the cabinet simulation - the amp PROFILE still includes the tube power amp simulation, which then runs through another physical tube power amp. Some people like the resulting sound, but it's at least not authentic. I would call this a work around setup.
I would recommend to aquire a KEMPER Power Kabinets instead, There are 3rd party alternatives, but the KEMPER Kabinet is the most flexible approach. You could add a second KEMPER Power Kabinet for stereo sound at any time.
