Not sure if it would even work but has anyone done a profile with a compressor in the signal path?

I think it would impact the Kemper's ability to accurately profile the amp. One of the various tests it does while profiling is how the amp reacts to volume and signal transients.

Well, compression IS something which happens in an amp anyway (if it is driven sufficiently)... i think it should work

Some channel strips used by some profilers introduce compression as well. Use sparsely and wisely.

I'm looking at something fairly transparent and obviously I know I can add compression after the fact but I'd rather get the tone I want in the profile. Ordered a Cali76 as I'd got rid of most of my pedals when I got the Kemper so once it arrives, I'll give it a bash and let you know how I get on!

There's a 1986-7 BK Butler Tube Driver in the chain too so believe me, I'm hoping the profile matches the tone that's coming out already without the compressor!

If you want the profile to match the feel and that is coming out without the compressor, I suggest making the profile without the compressor. Or, at least, make some without it, and some with it, and see which are more like the amp.
