Does anyone just profile an amp with all its settings at 12 o'clock and then adjust the Kemper EQ controls as needed?

Not me.
It would obviously be possible and might even provide some interesting sounds. However, mosts amps aren’t operating in their sweet spot with 12 o’clock settings. The mid settings on most tone stacks are far from flat as many people believe. In fact most tone stacks are highly interactive and have an effect on gain so I would always aim to get the amp sounding great then profile the result. -
I profile my amps in many configurations. For example, I profiled one amp more than 100 times, with different settings/cabs/speakers/mic placements.
